The South Sydney fullback will partner Manly's Tom Trbojevic in the centres.
Mitchell, who switched from the Sydney Roosters to South Sydney last year, returned from a four game suspension for South's comfortable win over Parramatta on Saturday afternoon
He was in great form before the suspension and led the Dally M leaderboard after the first seven rounds.
Mitchell was a member of the 2018 series winning NSW side, where he played three games in the centres. However, he was dropped after NSW lost the opening game in 2019. Mitchell missed last year's series because of injury.
"I'm really happy with the squad we've announced to face the Maroons for the opening game of the series," NSW coach Brad Fittler said.
"We've got a good mix of players from in-form teams and players who have worn the NSW Blues jumper with distinction in the past and never let their state down."
