Simmons stepped up and buried from outside the arc in the first quarter of the 109-104 win against New York Knicks on Wednesday.
He said in an on-court interview post game that he had been "repping" the shot in practice.
"It felt good ... I just like to come out here and play."
It was his first successful three in 194 NBA regular season and playoff games - an astonishing statistic for a modern day All-Star point forward with Simmons' talent.
Philadelphia's Wells Fargo Center exploded in jubilation, and social media caught fire with Simmons' shot the top trending topic on Twitter within minutes.
"It's happening," Sixers coach Brett Brown told reporters.
"He's 23-years-old ... this is a small step, but a step toward the other pieces of his growth.
"Other people get more excited than I do. But how can it not be a good thing? But if we're all honest, that's the first legitimate 3 that he's taken."
Simmons has only attempted 18 three-pointers in regular season and playoff games and missed all of them until Wednesday.
The Aussie had 18 points, seven rebounds, 13 assists, one steal and one block, and did not try another three-pointer.
The 76ers (9-5) overcame a 17-point third-quarter deficit to beat the Knicks (4-11) with Mike Scott and Joel Embiid hitting three-pointers in the final minutes to give the hosts the win.
Luka Doncic finished with 35 points, 11 assists and 10 rebounds as Dallas Mavericks routed the short-handed Golden State Warriors 142-94.
The second-year star from Slovenia became the first player in NBA history to record a 35-point triple-double in 25 minutes or less, coming off a 40-point triple-double on Monday night against San Antonio.
The Mavericks (9-5) tied a franchise record with 22 three-pointers while sending Golden State (3-13) to their worst loss since 1973.
