United will meet Wauchope at RSL while Wingham will clash with Port Panthers at Cedar Party Reserve.
Taree West will be on the road, taking on defending premiers Macquarie Hotel at Oxley Oval in Port Macquarie.
In the other match Nulla from Kempsey plays Port City Leagues at South Kempsey while another Kempsey club, Rovers, sits out the week with the bye.
The same nine teams that took part in the inaugural premier league last summer will play in 2018/19. Last season Wingham were finalists while United finished in third place.
Taree West failed to make the playoffs.
The competition will run for 18 rounds with the last matches on March 23. April 13 will be the date for the final at a ground to be decided.
Port Macquarie will host the Regional Big Bash T20 round on October 6 and 7.
