Police were called to the Pacific Highway at Moorland about 4.15pm after a southbound Toyota Echo and a northbound Toyota Tarago, which was towing a boat, collided.
The driver of the Echo, a male, died at the scene.
The male driver and female front seat passenger, as well as their four child passengers, were taken to Port Macquarie Hospital for treatment to minor injuries.
A contra flow is currently in place at the scene.
Police are urging drivers in the area to take care, with heavy rain and a large amount of water on the road.
Officers have attended several reports throughout the day of vehicles leaving the road as a result of the conditions.

5.30pm: A contra flow operation is about to be put in place on the Pacific Hwy near Valhaven Rd due to a serious two car crash.
Exercise caution and allow extra travel time through the area.
SUNDAY 5pm: All northbound lanes are closed on the Pacific Highway at Moorland due to a serious two-car crash near Valhaven Road.
Emergency services are on scene and traffic response crews are attending.
Motorists are advised to avoid the area, use an alternate route or delay northbound travel on the highway.
