Captain Brooke Murray, Ashleigh Green, Keely Holden and Taryn Dixon will miss the competition, but 13 of the 17 members of the premiership winning side are expected to play.
"We're down a couple, so we lose a bit of depth, but there's still a lot of good talent there," Jacklin said.
Sixteen of the best teams in New South Wales will battle it out to be crowned the State's number one league tag side in 2015.
St Johns Oval in Central Charlestown will host 27 matches across two fields with the winner taking home $3500 in prize money and the title of CRL Women's League Tag Premiers Challenge champions.
Jacklin said the team was interested in seeing the difference between other team's and their playing styles.
"It'll be interesting to see how other sides play and the structures they use," Jacklin said.
The challenge will consist of four pools of four teams, with each side playing each other once.
The majority of teams involved are premiers. Taree will have Ashford, Newcastle and the Entrance in their pool.
The highest ranked side from each pool will progress to the semi-finals, with the winners of each semi-final facing off in the grand final.
CRL chief executive Terry Quinn believes the challenge is a wonderful opportunity for the thousands of women who play league tag across the State.
"The growth of league tag over the last few years has been nothing short of exceptional and the Premiers Challenge is a great way to bring the best of the best together," Mr Quinn said.
The semi-finals and final will be broadcast live on Youtube, with the link available on the CRL website.
